Valorisation des fractions issues d’un procédé industriel éco-compatible de raffinage de macroalgues marines

Abstract : SEPROSYS company is specialized in the development of processes for the extraction, separation, and purification of molecules of interest. In 2011, the company developed an innovative and eco-compatible process for the separation and fractional purification of biomolecules from marine macroalgae. The aim of this thesis is to identify potentially applications for the different fractions extracted using the process developed by SEPROSYS, and in particular, in two distinct fields: in dermocosmetic for anti-aging and anti-stain activity, and in nutraceutical for anti-inflammatory activity. The first part concerns the improvement of the process, the characterization of protein fractions as well as the production of peptide and oligosaccharide fractions from Ulva rigida and Ulva intestinalis. The second part is devoted to the evaluation of the "anti-aging" and "anti-stain" potential of the different fractions from Ulva rigida and Ulva intestinalis. Thus, the impact of our fractions on the biosynthesis of collagen and hyaluronic acid from fibroblast lines were studies, as well as the effect of the fractions on the in vitro production of melanin by murine melanocytes. In addition, the aim of this work is also to better understand the structure-function relationship of the anti-aging activity of protein fractions. The third part concerns a project called Neuronalg, whose objective is the development of algo-sourced nutritional actives, targeting inflammation and mechanisms of alterations in emotional behaviour linked to metabolic disorders. This will be achieved through the anti-inflammatory activities of fractions on mouse macrophages through the quantification of nitric oxide produced, associated with the transcription level of target proteins such as iNOS (inducible NO synthase) and pro- and anti-inflammatory cytokines.
